چکیده

Abstract Study question In men with Raised Sperm DNA Fragmentation Index (DFI), Would advance sperm preparation techniques at Intra-Cytoplasmic Injection (ICSI) alter embryo euploidy status? Summary answer raised DFI, use of selection are safe and do not appear to ploidy status the embryos What is known already MACS (Magnetic Activated Cell Sorting), Microfluidic Sorting (MF), surgically retrieved Testicular (TESA) Double Density Gradient processing (DDG) have been shown optimize ICSI for DFI. any these methods proven design, size, duration This was a retrospective observational study conducted private fertility centre in-between years 2015 & 2022. Couples where women (<37yrs) had idiopathic Recurrent Implantation Failures (RIF) DFI (>25% DFI) underwent Pre-Implantation Genetic Testing (PGT) were evaluated (n = 473). Based on method population divided into four groups. TESA 84), 105), MF(n 135), DDG 148).A total 1802 in this study. Participants/materials, setting, All Controlled Ovarian Stimulation (COS) oocyte retrieval male partners as per clinics SOP. Appropriate counselling written consents obtained from all couples, applicable. oocytes subjected ICSI, extended blastocyst culture trophoectoderm (TE) biopsy. Post biopsy vitrified TE tissue Next-Generation Sequencing (NGS) assess status. Ploidy between various compared. Main results role chance Euploidy rates groups follows: 311 screened)- 48% 445 screened) – 51% MF 489 61% 557 54% Though numerically highest group euploid embryos, there seems be no statistical significance (p-value 0.973). Different seem Ejaculated or sperms similar Use sperm, which considered immature doesn’t increase aneuploidy. MACS, seemed embryos. further warrants research validate routine intervention reproductive outcomes. Limitations, reasons caution Retrospective un-equal size small sample well-designed needed findings Wider implications Role implantation yet proven. Data showed alarming aneuploidy younger comparable published literature. Trial registration number applicable

The effect of rhFSH on Sperm DNA Fragmentation and sperm parameters in Oligozoospermia Infertile Men

Background: One of the main causes of male infertility is the negative effects of oxidative stress. Follicle-stimulating hormone (FSH) plays an essential role in spermatogenesis, as well as in the maintenance of sperm DNA integrity. This study aimed to determine whether the recombinant human follicle-stimulating hormone (rhFSH) treatment of sperm parameters could positively affect sperm DNA and...

P-193: Chlamydia Trachomatis Can Increase Sperm DNA Fragmentation in Infertile Men

Background: Chlamydia Trachomatis (CT) is an obligate intracellular bacteria, that requires living cells to replicate itself and is responsible for lower urinary tract disease in men and women. It can cause urethritis, epididymitis, orchitis and complications such as chronic prostatitis in men. Chlamydia infections may also directly affect male fertility by damaging the sperm, because sperm par...

P-240: Effect of Oxidative Stress on DNA Fragmentation Index in Sperm of Mature Mice

Background: One of the major factors affecting on male infertility is oxidative stress (OS). Two main sources of ROS production are leukocytes and spermatozoa. The ROS such as tertiary-butyl hydroperoxide (TBHP) may damage and fragmentate to sperm DNA, which can lead to complications in the offspring. In this study, we will investigate the effect of od on DNA fragmentation index (DFI) in sperm ...
